Sounds like the INI is pointing to a non-existent path. That might happen when, for example, a USB (or other mountable drive) is removed that once was the path for input or output.
Try deleting BDREBUILDER.INI (from the installation folder) before starting BD-RB. It will re-create it with default settings.
